    What I think more likely is that the acid-base extraction doesn't favor any particular alkaloid (I could find no reason to suspect it might) and the explanation for the different alkaloid profile is that in smoking, the 'MAOI' alkaloids (or the 'others') are less readily evaporated than nicotine...

    It would be good to determine if / to what extent the grinding of the tobacco is important for yield - perhaps less than one would think. Perhaps extend the period to compensate. With the hot oil approach in particular, maybe the fine grind is unnecessary?

    Either the more volatile (?) non-polar sol vent is better at dissolving non-alkaloids (in which case the color would largely disappear on the second stage) or, more likely, the darker color is from the alkaloids being much more oxidised in this second run (and 5 day steep).

    This is a great innovation. I think your complete method, with all it's tips and carefully devised honing of the acid-base extraction method to the WTA purpose, is about as good as a home method can get - i.e. not much short of what a lab could do.
