Only if you partially block them. If you were to completely block them it would be much like sucking a baseball through a slurpee straw. I accidentally covered them doing a ghetto PV repair with electrical tape and couldn't draw on it at all.

You need to leave at least one open to get some air in it or your PV won't work.

You can easily block them with your fingers and see that no air gets through.

I've seen people use scotch tape to block one or more to adjust air flow. I had some "airy" cartomisers so I ran a piece of tape around the carto at the point it exited from my cone to restrict air that way I liked that method because I didnt have to take it off to change cartos like you do if you use tape to block the air hole directly.

If you are not using the cone, use the cone or break a piece of toothpick in one of the slits.

If you're using cartos, you can turn the small slot on the bottom 1/4 turn in (to the right) with a small screwdriver to tighten the draw. Turning it more than 1/4 inch can do damage.

If you have any carto condoms, cut that and use it to partial cover.

I have a new eGo with cone. I was using a clearomizer with a slight leak so I wound a small piece of cotton around the bottom where it was leaking, then pulled the cone down, tucked the cotton under, and screwed the cone down. Not sure if it is advisable, but it did tighten up the draw, it also kept things from getting overly juicy.
