It is upon the very first filling or whenever a tank and carto are dry. By the time you filled your tank the carto is covered in the juice so it's pretty well lubed up, it should just slide right up into the top cap opening.
One thing you might consider trying is swapping the o-rings between the two caps so that the bottom is nice and snug and the top is a bit looser. This might help prevent leaks unless you like vaping upside down.

Thanks Snow, you've set my mind at rest. I've used a gold pen to paint around the flange seating, and this time when I refilled, I could easily see where to fit it back to. And I didn't pop the top cap off. I'm getting better at it, in fact I'm getting rather too much practise. Maybe I need to go for a 6ml tank - at least for my Nuport?

Just last night I was lying on the deck, vaping & looking up at the Milky Way through binoculars. On a warm summer evening, it is amazing. Perhaps instead of buying all this new vaping gear, I should buy myself a decent telescope so I can better see into those tantalizingly fuzzy areas between the stars. Maybe start seeing nebulae, and all those tempting morsels of the night sky.
