I'm using the V2 ex-blanks. It says they can be refilled about 15-20 times but that it might be less if not using their own ejuice, which I'm not, so how do I tell when it needs replaced? And, am I meant to let it run out completely before refilling it again? Is it bad to just keep topping it up?

Also, is it normal for the ejuice to look darker as the tank runs out? Mine definitely looks darker than the brand new ejuice.

1-Never run your tank dry, this will burn the cotton/coil. Time then for a new coil.
2-Topping the tank will hurt nothing, see #1.
3-If your juice is getting dark, I would suspect that #1 has occurred.
4-The cartridge is optimized for use with high pg eliquid and is not compatible with high vg liquid.

Yes, I usually don't drain my tanks far past about 1/4 of the liquid. I usually run different flavored profiles in different tanks. It is possible your juice will darken after repeated refills, regardless if you do it sooner or later. The coil life you will get is also dependent on the type of juice you use. Dark, sweet, and NET (naturally extracted tobaccos) will burn through coils quicker than less sweet, clearer juices) but it is totally normal to have juice darken in ones tank as it is heated and vaped. Some sweeteners (or too much) can caramelize on the coil without burning it completely, so it's not unusual to see juices grow darker as you near the end of your coil's useful life.

V2ex uses tiny little pieces of silica wick not cotton.
To extend their life use a higher Pg liquid, they were designed to use 80pg /20vg, they'll work fine using 50/50 but it will shorten their life slightly.
Replace them when the draw tightens up, flavor goes funky or produces less vapor than usual or stop working.
It is normal for the liquid to darken when using the tiny bottom coil clearo's, you can kinda clean them out with PGA or vodka, fill about half way, shake vigorously let sit a few minutes shake again, dump out, rinse in hot water, allow to fully dry, refill.
It is better to keep them filled at least enough to keep the bottom coil & wicking covered in liquid.

I agree with Topwater Elvis's information.

To answer your specific questions:

You can add more e-liquid at any point in time. You don't have to wait until the tank is almost empty, but you don't want to let the tank go dry, as that will cause what's known as a "dry hit" -- totally nasty.

E-liquid will naturally turn darker over time. Exposure to oxygen causes the nicotine to turn dark (not a problem). Unvaporized components in the e-liquid (flavoring agents and sweeteners) will progressively accumulate in the tank and give it a darker appearance; not harmful, but will probably negatively affect the taste.

You're using what is known as "old technology". You could get a better vape if you upgrade to a newer tech clearomizer. Proper Terminology: Is it a carto, a tank, or what? A Guide to Juice Delivery Devices
