Okay, so I don't really know how to ask this question but I will try my best. I'm sorry for any confusion.

E-cigarettes use Water, Propylene Glycol (or Vegetable Glycol), and Flavoring.

How do the companies make the flavoring for the cartridges? what is the flavoring made of?

The ejuice that is put in prefilled cartridges is the same make up of the ejuice you buy in bottles. The flavoring used in creating ejuices is mainly in a PG base with the flavor extracts added. The extracts should be (and is in most cases) not oil or sugar based - as those are bad for atomizers - so most are labeled as 'water soluable'. There is a chart for the lorann flavors here:
