They don't ferment. If you're mixing with a high proportion of VG then some time could be allowed for the flavour to "steep" or soak through into the entire liquid. Since most people use 70% PG or higher this usually isn't necessary.
I always try it right away, and sometimes it is good enough to vape. If you let it steep, the flavors seem to blend and deepen, so if it doesn't seem flavorful enough I let it rest for a while. A day with the cap off, in a nice dark place. Replace cap and shake daily and keep trying it.
I put my juice in a glass amber bottle, cap it, shake the crap out of it, then I put a bowl of water in the microwave for 3 minutes (don't let it get to boiling). I take off the cap, put the bottle in, and leave it until the water is room temperature. I've been doing this since I've been doing DIY and all of my creations have blended pretty well so I can taste them about 30 minutes after making them.
Like others have said, it depends on the PG/VG ratio. I use higher VG than most, so I let my juices steep for about two weeks, uncapped for the first two days.
