If your speaking about cartomizers. Yes, the polyfill is in there to absorb the e-liquid that you put in there. If your using standard sized cartomizers (EX. Boge LR 2.0) than it will take 25 drops. Let it seep for 20 minutes before using to ensure that everything is wet.

If your using XL cartos (Smoketech Dual Coil Cartomizers) than 45 drops is the max.

After the initial fill than you want to top it off or add additional 5 drops after 15 hits or so.

thank you :) another question, the cotton inside is a filter for the liquid right? and how much dosage of the liquid should I put to refill everytime?

Not so much a filter, more of a reservoir/wick that holds the liquid and feeds it to the coil for atomizing.
As others have said generally keep it wet, it will take some time and trial and error to recognize when it needs to be re-wet depending on how much you vape. There are some good links on here as to refilling tecniques.
