Is this a cartridge aka cart which is used with an atomizer? I never got any prefilled 510 carts but they have a rectangular juice cup and you should try to orient it with the atty bridge this way

If you mean the actual mouth end with the air hole came off - wow no clue - sorry.

Or do you mean a cartomizer aka carto which screws on to the battery? And if so what kind?

I cannot picture what you ware talking about. CE2, Clearomizer, Fluxomiser cartos have an inner seal (possibly 2) and you are supposed to use a special syringe (I think with a flexible tip) to fill them through a hole or port in the seal under the mouth cap. If you removed that inner seal, the orientation is with the side notches fit on something in there.

I'm talking about a white soft silicone plug that is inside the mouth end of a 510 pre-filled cartridge.
This white silicone plug is pan-shaped it has a hole in the center with a extruding tube. It is located inside the cart's mouth end under the hard plastic press-on end cap. I snapped the caps off a couple other carts and the silicone plugs were haphazardly oriented. Common sense tells me the pan bottom should be oriented towards the mouth piece cap but I'm not absolutely certain.

That ring should be set like a plug-not sideways. It will catch e-liquid if you overfill a bit and it attempts to keep the liquid from getting in your mouth. Seeing as the area it is in is very small it won't catch a whole lot. If your overfill is a lot then you will get a bit of juice in your mouth. Remember to remove that plug and get out any extra juice that may get in there when you over fill. If you look closely the ring should have a groove in it, that groove should be facing towards the filling end of the cart not the end you put in your mouth, it helps to hold overflow.

Edit-I have never seen one with the extruding tube. They must be trying to improve on them. If you can see which end may help hold any juice, put that end opposite from the end you put in your mouth.
Lordy, I hope this makes sense. LOL
