Good old Ohm's law shows the difference. The actual power available is slightly higher, as in about a watt and a half, with stacked 18350s versus a single 18650. Plus, when using a single 18650 at higher voltages (even effective voltage in RMS mode) the current demand rises to achieve the higher voltages over 3.7 volts of the battery.

There should be no difference because the voltage is regulated right to cut off. However, tests did show the Vamo hits a little bit harder in watts mode with 2x 18350. And you can go up to 15 watts with 2x 18350.

That was on a version 1 Vamo, but its the same chip as version 2.
