Okay, so if I turn the charged up battery on -- the light blinks 5 times and it's ready to use. If I leave it turned on, will that run the battery down just because it's on, regardless whether or not I'm actually vaping?

The clicking is locking/unlocking the button functionality. IT IS A SAFETY FEATURE ONLY.
The battery is only "on" when you press the button to use it.

I only use the off feature when it is in my pocket - removes the chance of it firing up in my pocket.
My Twists & Spinners never leave the house.
Since I use my Blu 100 when out.

There is no need to turn it off unless it is going to a place where the button could get pushed unintentionally, like a pocket/purse/drawer...
I never turn mine off unless it's going to be in my pocket for an extended amount of time. Now, this is just my opinion here, but seems to me that constantly clicking the button 5 times (the LED lights up everytime) to turn it on and off would wear down the battery enough to negate what battery life might be saved by turning it off.
I turn mine off every time I set it down, stick it in my pocket, or put it in my purse... for different reasons. 
My PV is usually an arm's reach away from wherever I am. If I didn't turn it off, it would likely end up in between couch cushions or wedged underneath my pillow. That would not be good.
I think it wouldn't make much difference in battery life if you left it on... even turning it off shouldn't take too much extra power, the led isn't going to be a major drain on the battery, I would think.

I just bought a great leather eGo case from Johnson Creek.
It's small & has a great belt clip. Twists & Spinners are a tight fit.
I will finally shut them off, when they're inside the case!

I turn my twist off just because my parents drilled into me, "If you're not using (insert electronic), turn it off!" Does it make a difference? Probably not.
I never really use that feature unless it's in my pocket or backpack with other things. It's not really an 'off' or 'on' thing cause it's only a lock. As stated before, your eGo is only 'on' when the button is depressed. I guess in effect it's keeping it in an 'off' position.
I only turn mine off at nite when I go to bed or when I am out and about putting in my pocket doesn't run down like others said just a safety feature so it doesn't come on in your pocket or purse
