So nic strength is 1mg/1ml right. So if I have using the equation above 1/4 12mg juice and 3/4 6mg juice (did it backwards before. Then by my math that's 4.5mg of nicotine but if my tank is 4.5ml then don't I have to multiply the 2 to get the full nic content in the tank.

No. You are working with liquid with X milligrams per milliliter. It's all fractions/ratios when mixing.
If you made 4.5mg/ml juice and filled a 4.5ml tank, you'd have 20.25mg total nicotine in the 4.5ml of juice in your full tank.
