Yes. There will be a slight changeover period where the flavors blend but eventually your new flavor will take over. That is unless your older flavor is super powerful. Worst case give it a rinse with warm/hot tap water, then reinstall it with your new flavor. It will be really weak until the water evaporates but will then pick up to normal.

It will also depend on the juice. I ran a tank of Suicide Bunny's Madrina through my subtank and it took soaking the rubber seals in rubbing alcohol to get the taste of melon out of them. They kept "flavoring" the new juice. I still have a bit of it showing up in the straight menthol I have in the tank right now, but I figured out that it is the seal in the top of the RBA chimney that absorbed the flavor as well. I'll soak that the next time I break it down for a wick change.
