The point is to get more eliquid to the wicks and hopefully help avoid dry hits. If it is a new coil, or a freshly cleaned coil, I do maybe 2 or 2 primer puffs. Otherwise, no, I do not do primer puffs.

What BigCatDaddy said^^ When I rebuild I use cotton wicks so I will put a little juice on the cotton before installing the chimney. If just refilling, no priming, just load and go!
Depends on the tank, if its a top coil or bottom coil, or a new coil and wick. It it's a RBA I do primer puffs on a new build, and same with top coils. The only bottom coil tanks I do primer puffs on is a nautilus or a kayfun if I top fill it.
